About 29 people were detained by the Ghana Police Service on Tuesday in connection with the Arise Ghana event.
Police and protesters exchanged stones and tear gas during a standoff in the afternoon as the protest descended into chaos.
Police accused the protesters of trying to take prohibited routes as they were protesting the country's escalating economic distress.
The organisation stated that they were being prevented from speaking out against the Akufo-Addo-led administration by the state security agency.
The immediate release of the protesters was previously asked by the attorneys for the 29 persons who had been detained on the first day of Arise Ghana's Krom Ay3 Shi march.
On the first day after their detention, according to their attorneys, they were refused access to their clients.
Abraham Amaliba, the protesters' lead attorney, had previously pleaded with the police to free his clients on a police investigation bond because there would be no court proceedings on Thursday in observance of Martyrs Day.
"Unfortunately, the courts are not in session today. The courts will be closed today in observance of or celebration of Martyrs Day, leaving the Police with little choice except to release the suspects into the custody of the police.
